Objective: Many nutritionists recommend that consumption of herbal derivatives could provide antioxidants compounds that are necessary for the human body. In this research, effects of aqueous extract of Iranian white tea (clone 100) were tested in colorectal cancer cell line HCT-116. Material and Methods: Different concentrations of white tea extract (WTE, 100-1000 μg/ml) incubated for three time periods (8, 16 and 24 hr) were examined by MTT assay for growth inhibitory effects in cells. Fold-change of protooncogenes and oncogenes (MMP2, K-ras and β-Catenin) was estimated by using real time-PCR assay and compared to the expression of Beta-actin as a housekeeping gene. Results: MTT assay result showed that three concentrations (150, 500 and 1000 µg/ml) of white tea extract, had a significant inhibitory effect on the growth of colorectal cancer cells. Treatment of cells with selective concentrations of WTE was accomplished for 8, 16, and 24 hr. Treatment of cells with 150, 500 and 1000 μg/ml of WTE for 24 hr, significantly decreased expression of MMP2. as Also, expression level of K-ras in significantly decreased by all three concentrations of WTE in 24 hr. Also the expression level of β-catenin after 24 hr, was significantly decreased by all WTE concentrations Conclusion: It seems 1000 μg/ml white tea extract in 24 h play an effective role in cell cycle of colorectal cancer according to IC50 result and significantly reduced fold-change of all study samples gene
